Identification of biomarkers to predict safety and efficacy of cow’s milk oral immunotherapy by peptide microarray

چکیده
Methods We studied 25 CM-allergic children at baseline, after oral desensitization, and at 6, 12 and 24 months of follow-up. Seven patients who refused CM-OIT were used as controls. Patients were classified as low, moderate, and high risk according to the number of allergic reactions (safety), time required to achieve tolerance (efficacy), and requirement of premedication. IgE and IgG4 peptide microarray immunoassay was performed using a library of overlapping peptides of CM proteins.

but robust approach towards the identification of markers that enable the prediction of the outcome of oral immunotherapy for cow’s milk allergy. By means of ELISA, the authors determined specific IgE, IgA, IgG subclass levels against cow’s milk extract, natural single cow’s milk allergens and the egg allergen ovalbumin in 40 children with cow’s milk allergy before and after immunotherapy. Seve...
متن کاملPD28 - Is other bovid mammals milk tolerated by children who have been submitted to cow’s milk oral immunotherapy?
Introduction Caseins cross-reactivity rises up to 90% between cow’s milk (CM) and other bovid milks. There are few studies on tolerance to other mammals milk in patients submitted to cow’s milk oral immunotherapy (CMOIT). AIMS: To determine the percentage of patients whom after CMOIT tolerate water buffalo (WB), goat and sheep’s milk proteins (MP). To describe immunologic features which can pre...
متن کامل“Allergy to Goat/Sheep’s Milk with Good Tolerance to Cow’s Milk but Not to Cow’s Milk Cheese: Identification of 1-105 Peptide from κ-Casein as the Molecular Basis of Reactivity toward Cow’s Milk Cheese
In Europe, a new population of patients developing anaphylaxis to goat’s/sheep’s milk associated to tolerance to cow’s milk has recently appeared. Nevertheless, among these patients, some show allergic reactions to cow’s milk cheeses but these reactions are less severe than those directed against goat’s/sheep’s milk. The objective of the study was to the molecular determinants of the reactivity...
متن کاملIn silico identification of epitopes from house cat and dog proteins as peptide immunotherapy candidates based on human leukocyte antigen binding affinity
The objective of this descriptive study was to determine Felis domesticus (cat) and Canis familiaris (dog) protein epitopes that bind strongly to selected HLA class II alleles to identify synthetic vaccine candidate epitopes and to identify individuals/populations who are likely to respond to vaccines. FASTA amino acid sequences of experimentally validated allergenic proteins of house cat and d...

AIM In cancer immunotherapy, biomarkers are important for identification of responsive patients. This study was aimed to find biomarkers that predict clinical outcome of WT1 peptide vaccination. MATERIALS & METHODS Candidate genes that were expressed differentially between long- and short-term survivors were identified by cDNA microarray analysis of peripheral blood mononuclear cells that wer...
